چکیده

Pseudomembranouscolitis is one of the serious infectious diseases with a high mortality. It tends to occur in association with antibiotic therapy, especially with broad spectrum antibacterial agents. The major causative pathogen is the anaerobic bacterium Clostridium difficileP This bacterium is extremely sensitive to oxygen, and requires strict anaerobic conditions for growth. Once exposed to air, it cannot grow any further. Therefore, all processes for the isolation, incubation for growth, and preservation of the bacterium have to be carried out in anaerobic boxes or anaerobic chambers. This requirement offers considerable inconvenience in the development and running of screening systems for agents effective to control pseudomembranous colitis. Wedescribe here a simple and convenient agar medium for growth of C. difficile, which can be handled under air for several hours, followed

Clostridium perfringens. I. Sporulation in a biphasic glucose-ion-exchange resin medium.

A biphasic culture medium suitable for cultivation and sporulation of Clostridium perfringens, C. botulinum, and C. sporogenes was devised. The medium designed for use in a disposable, compartmented, plastic film container contained peptones, yeast extract, minerals, an anion exchange resin, and glucose in 4% agar as the solid phase and (NH(4))(2)SO(4) and 0.1% agar as the liquid phase. With th...

New medium for rapid screening and enumeration of Clostridium perfringens in foods.

A rapid and sensitive procedure for estimating low numbers of Clostridium perfringens has been investigated and compared to methods used currently in the food industry. The new liquid medium, RPM (rapid perfringens medium), was compared with sulfite-polymyxin-sulfadiazine agar and tryptose-sulfite-cycloserine agar in recovery studies with naturally contaminated and with inoculated foods. The me...

Effect of L-cysteine on the activity of penicillin antibiotics against Clostridium difficile.

We observed elevated MICs of penicillin antibiotics while performing agar dilution susceptibility testing of strains of Clostridium difficile on supplemented brain heart infusion agar, an effect which was completely eliminated by the exclusion of L-cysteine from the medium. L-Cysteine antagonizes the activity of penicillins against C. difficile, most likely by direct inactivation of the antibio...

Effect of inoculum, pH, and medium on the activity of ciprofloxacin against anaerobic bacteria.

The in vitro activity of ciprofloxacin against 30 Bacteroides fragilis group, 30 Clostridium, and 30 Peptococcaceae strains was determined by the agar dilution method in two different culture media at three pH and in three inoculum densities. In Wilkins-Chalgren agar the MICs for 90% of the strains were 0.12 micrograms/ml for B. fragilis, 0.5 micrograms/ml for Clostridium spp., and 2 micrograms...

Selective and differential medium for isolation of Clostridium difficile.

Clostridium difficile is a recognized cause of pseudomembranous (antimicrobial agent-associated) colitis and may be one of the causes of antimicrobial agent-induced diarrhea. A selective and differential agar medium that contains cycloserine, cefoxitin, fructose, and egg yolk (CCFA) was developed to facilitate the isolation of C. difficile from fecal specimens. Quantitative cultures of 16 stock...
